Protein A is a cell wall protein of Staphylococcus aureus that can specifically bind to many mammalian IgG and a few IgM and IgA, mainly to the Fc region of the immunoglobulin heavy chain. Protein A can couple with various reporter molecules (fluorescent dyes, enzyme labeling, biotin, colloidal gold, radioactive labeling, etc.) without affecting its binding activity with antibodies. These coupling reactions can be used for antibody detection in ELISA tests, histochemistry, Western Blot, etc. Protein A can also be combined with agarose gel to prepare an affinity filler for antibody purification, which can separate and purify antibodies of various mammalian subtypes or genetic engineering recombinant proteins containing antibody Fc fragments from ascites, serum, cell culture supernatant or cell extracts.
